Fibrillarin is an excellent marker for the nucleolus. As a component of a nucleolar small nuclear ribonucleoprotein (snRNP) particle, it is thought to participate in the first step in processing preribosomal RNA. Fibrillarin protein contains an amino-terminal domain that is rich in glycine and arginine residues (termed the GAR domain), a central RNA-binding domain comprising an RNP-2 like consensus sequence, and a C-terminal alpha-helical domain. Fibrillarin binds to U3, U8, and U13 small nuclear RNAs. It associates with several other structural proteins as well as box C/D snoRNA to form a complex that functions in pre-rRNA processing, pre-rRNA methylation and ribosome assembly. This complex catalyzes site-specific 2-O-ribose methylation of targeted nucleotides within the rRNA sequence.
