Background |
The H1 histamine receptor is expressed primarily in the lungs, vasculature, and brain. The H1 mediates the contraction of smooth muscles, neurotransmission in the central nervous system, the release of catecholamine from adrenal medulla, and increases in capillary permeability due to contraction of terminal venules. H1's role in inflammatory responses makes its antagonist suitable for treating allergies. |

Figure 1. Histamine-induced concentration-dependent stimulation of intracellular calcium mobilization in HEK293/H1 cells. The cells were loaded with Calcium-4 (Cat. No. R8141; Molecular Devices) prior to being stimulated with agonist Histamine. The intracellular calcium change was normalized and measured by FLIPR. The relative fluorescent units (RFU) were normalized and plotted against the log of the cumulative doses of Histamine (Mean ± SEM, n = 3). The EC50 of Histamine on this cell was 0.65 nM.

EC50 value is calculated with four parameter logistic equation:

X is the logarithm of concentration. Y is the response
Y is % stimulation of RFU and starts at Bottom and goes to Top with a sigmoid shape.
