The surface expression of CD32B 232Ile is validated by FACS analysis. This stable cell line product is designed for measuring binding affinity and stability of Fc region of antibodies and Fc fusion proteins, or antibody based biologics binding with CD32B 232Ile. Background |
Human CD32 group consists of FcγRIIa, b and c proteins that share 94-99% amino acid identity in their extracellular domains but differ substantially in their transmembrane and cytoplasmic domains. CD32B is a low affinity receptor that recognizes immunoglobulin gamma complexes. It is expressed on B cells and most myeloid cells. CD32B has an intrinsic cytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM) and delivers an inhibitory signal upon ligand binding. Co-ligation of CD32B with B cells antigen receptor (BCR) results in selective dephosphorylation of CD19 tyrosine, tyrosine phosphorylation of the ITIM, and recruitment of SH2-containing inositol 5-phosphatase (SHIP). This causes a block in calcium influx, inhibition of B cell activation, and leads to apoptosis. CD32B is a potential target for monoclonal antibody therapy for malignancies. Targeting CD32B may represent a novel approach to immunotherapy of B-cell malignancies. |
