PCR Cloning and Subcloning TEM-1 was purchased from Invitrogen (Carlsbad, CA, PV3575). CTX-M-15 was purified by 45 Evotec Ltd (Abingdon, UK) after cloning into pET26-b(+) and expression in E. coli. CTX-M-14 46 was expressed and purified at GenScript (Piscataway, NJ). Ceftolozane and tazobactam were 47 supplied by Cubist Pharmaceuticals (Lexington, MA). Clavulanic acid was from Fluka/Sigma- 48 Aldrich (St. Louis, MO). Piperacillin, ceftazidime, levofloxacin, and tobramycin were from 49 Sigma-Aldrich (St. Louis, MO). Sulbactam, cefepime, and meropenem were from U.S. 50 Pharmacopeial Convention (Rockville, MD). Ceftolozane-tazobactam Etest strips were from 51 bioMérieux (Marcy-l’Étoile, France; lot# 1001256360). Ceftolozane MIC50/MIC90 values were 4/8 μg/ml when tested against 26 CTX-M-14-type- 25 producing isolates, and 64/>64 μg/ml against 219 CTX-M-15-type-producing isolates. Addition 26 of 4 μg/ml tazobactam lowered ceftolozane MIC50/MIC90 values to ≤0.25/0.5 μg/ml by broth 27 microdilution and Etest. Zone diameters for a ceftolozane-tazobactam disk were 23-29 mm for 28 92.2% of the isolates.
